Cell-mediated immune responses were examined in 36 healthy adult controls and 62 healthy elderly persons, and the following results were obtained. 1) Delayed hypersensitivity responses to two skin test antigens, PPD and Candida were significantly reduced in the aged than in the healthy adult. 2) The aged showed a significant reduction of the total number of lymphocytes in the peripheral blood when compared with those in normal adult control. 3) The subpopulation of lymphocytes was estimated by the sheep red cell rosette forming techniques. The proportion of T lymphocytes of the aged were significantly lower than that of the adult control, whereas the proportion of B lymphocytes showed no difference between the two groups. When the absolute numbers (counts per mm3) of both T and B lymphocytes were determined, counts of both lymphocytes were found to decrease significantly in the elderly persons as compared with normal adult controls. 4) The responses of peripheral lymphocytes to Phytohemagglutinin (PHA), expressed as the stimulation index (SI), were lower in the aged than in the adult control. 5) Negative skin tests to both PPD and Candida antigens were observed more frequently in the aged group showing decreased T lymphocyte counts as compared with the aged group with normal T lymphocyte counts. 6) There was a significant positive correlation between changes of T lymphocyte counts and of SI to PHA in the entire group of healthy adults and elderly persons. From these results, it is noted that the incidence of abnormalities in T lymphocyte function may increase by the aging process.
