Contractions induced by KCl or other solutions in briefly glycerinated thin bundles which were isolated from frog sartorius or semitendinosus muscles were studied and the following results were obtained. 1) When thin bundles were immersed in glycerol solution, an initial and secondary contractions occurred. The first contraction appeared immediately after immersion of the thin bundle in glycerol solution. The second contraction was seen after a distinct lag (7-8 min) and was superimposed on the first contraction. The tension of the second contraction was larger than that of the first contraction. 2) Both the first and the second contractions were observed at glycerol concentrations between 10 to 50%. The second contraction made its appearence only at higher glycerol concentrations ranging from 30 to 50%. 3) When briefly glycerinated thin bundles were immersed in a solution containing 0.15 M KCl, it produced a marked contraction which was followed by spontaneous relaxation. The peak tension of the contraction decreased and the time course of the contraction was prolonged with the increase of glycerinated period. 4) A marked contraction similar to KCl-induced contraction was elicited by immersion of the briefly glycerinated thin bundles in normal Ringer solution, distilled water or 20% glycerol solution. The contraction induced by normal Ringer solution was followed by spontaneous relaxation, while a marked spontaneous relaxation was not observed in the contractions caused by distilled water or 20% glycerol solution. 5) Both the peak tensions of KCl-induced contraction in briefly glycerinated thin bundles and K-contracture in intact thin bundles were inhibited at low temperature (0℃). The extent of inhibition in the former was larger than that in the latter. On the basis of these results, it was suggested that the KCl-induced contraction in briefly glycerinated thin bundles was caused by a direct interaction between residual ATP, ions and the actomyosin system within the muscle fiber.
