Changes in rat red cell shape as well as in lipid compositions were investigated after ligation of the rat bile duct.At the 12th hour after the ligation, spur cells were found to appear prominently, and continued to do so until the 4th to 5th day, after which target cells then appeared. Whereas in plasma the amount of phosphatidylcholine and cholesterol contents increased immediately after the ligation, such a rapid increase could not be found in red cells. Uptake of cholesterol into the erythrocytes clearly started at the 3rd day and reached its maximum level at the 7th day. The uptake of phosphatidylcholine into erythrocytes increased gradually and reached a plateau at about 5th day. The accumulation of these lipids in erythrocytes reflected the shape of the erythrocytes, e. g. target cells. Fatty acid compositions in phosphatidylcholine from erythrocytes changed in accordance with those in plasma. The changes in fatty acid composition in plasma phosphatidylcholine increased by the ligation seemed to be affected by those in bile. The present study made it clear that biliary obstruction brought about rapid changes in plasma lipid composition as well as changes in red cell shapes.
