Fifty-four cases of human colorectal carcinoma, 5 cases of familial polyposis of the colon, 13 cases of adenoma and 40 cases of normal colonic mucosa (30 adults, 8 neonates, 2 fetuses) were investigated in terms of gamma glutamyl transpeptidase (GGT) histochemistry and biochemistry as well as for the expression of mucosubstances. GGT activity was shown by histochemistry to be high and positive in the fetal and neonatal mucosa as well as in the adenomatous and cancerous tissues, while normal adult colonic mucosa was almost devoid of such activity. This was confined by the biochemical examination of the respective tissues. The distribution of mucin indicated the difference among the cells within the crypt. The cells of the lower crypt were composed mainly of immature cells and predominantly sulphomucin whereas those in the upper crypt were comprized of mature cells and predominantly possessed sialomucin, suggesting a correlation between the difference in the mucin and cellular differentiation. Adenomatous tissues invariably contained various types of mucin. However, the more dysplstic changes proceeded in the adenomas, the higher was the sulphomucin content observed. The mucin production by the cancer cells decreased significantly as compared with that of normal mucosa.
