The process of transportation of very low density lipoprotein (VLDL) particles in rat hepatocytes was studied electron microscopically utilizing cytochemical markers specific for the endoplasmic reticulum (glucose-6-phosphatase), Golgi apparatus (thiamine pyrophosphatase), and lysosomes and GERL (acid phosphatase). A considerable number of VLDL particles were seen in the hepatocytes and in the space of Disse, when the rats were allowed free access to 10% sucrose in drinking water for 5 consecutive days. They were frequently found in the terminal portions of rough endoplasmic reticulum and in the anastomosing channels of smooth endoplasmic reticulum near the space of Disse, suggesting that VLDL particles are formed in the cisternae of rough endoplasmic reticulum and then transported through the smooth endoplasmic reticulum. Cytoplasmic microtubules were frequently found in close association with the endoplasmic reticulum. When a low dose of colchicine was injected into the sucrose-treated rats, the secretion of VLDL particles into the space of Disse decreased markedly. VLDL particles were found in the cisternae of GERL. Few microtubules were found in the hepatocytes after colchicine treatment. The significance of microtubules in the transport of VLDL particles in the hepatocytes was discussed.
