Xenoantiserum highly specific for rat Thy-1 antigen (ABS) was prepared by immunizing rabbits with partially purified rat brain Thy-1 antigen. This antiserum reacted with majority of rat thymocytes, approximately 30% of bone marrow cells and 10% of spleen cells. When anatomical localization of ABS-reacted cells within the rat thymus was examined by immunoperoxidase staining, thymocytes located in the thymic cortex were intensely stained. Electron microscopic observations were made on bone morrow cells, and Thy-1 antigen was detectable on lymphocytic and immature blast cells, but not on mature nucleated erythrocytes and granulocytes containing peroxidase-positive cytoplasmic granules. We used the radioimmunoprecipitation technique and reacted ABS with various rat lymphoid cell lysates that had been labeled by sodium 3H-borohydride after periodate oxidation and by lactoperoxidase catalyzing 125I-iodination. SDS-PAGE analysis of the immunoprecipitates showed that ABS reacted with a 24,000-27,000mol.wt. component from rat thymus and bone marrow cell lysates, that could be bound to a Sepharose-4B column coupled with Lens culinaris hemagglutinin and that could be labeled by sodium 3H-borohydride, suggesting that it is a sialoglycoprotein. The immunological function of Thy-1 positive spleen cells was investigated and it was found that Thy-1 was expressed on NK cells as well as on precursor cells of Con A activated suppressor cells but not on cells responding in a allogeneic mixed lymphocyte interaction.
