Animal experiments were carried out using rats on the inhibitory effects of secretin, H2 receptor antagonists and other drugs, in the presence of gastric stimulation with tetragastrin, histamine and insulin in association with the regulation mechanism of gastric acid secretion. 1. Continuous i.v. administration of tetragastrin or histamine caused constant acid out put in rat gastric secretion; the stimulation was found to be dose dependent. 2. One-shot adimnistration of secretin or H2 receptor antagonists (metiamide and cimetidine) showed inhibitory effects on the continuous gastric acid secretion in the presence of administered tetragastrin or histamine. 3. The inhibitory effect caused by secretin showed a competitive type on Lineweaver-Burk equation of gastric acid secretion in the stimulation with tetragastrin, and also showed a competitive type of inhibition in histamine. 4. The inhibitory effect caused by cimetidine showed a competitive type on Lineweaver-Burk equation of gastric acid seretion in the stimulation with histamine, but did not appear to be competitive in tetragastrin.
