Hepatoma was prepared by the administration of 3'-Me-DAB to the rats in order to study the pathophysiology of experimental carcinomatous neuropathy, which was quantitatively analyzed by nerve conduction velocity of dorsal nerve trunks with an assessment of histological examination of the sciatic nerve and was also simultaneously investigated in terms of folate metabolism. 1. The motor nerve conduction velocity, which was markedly delayed from the early stage of hepatoma, did not show a close correlation with the progress of hepatoma. 2. The teased fiber of the sciatic nerve demonstrated degenerative changes of myelin sheath at the paranodal area of Ranvier reflecting delayed nerve conduction velocity. 3. The hepatoma rats revealed low serum folate levels, and the folate concentration fell in accordance with hepatoma progress in the liver, spleen, kidney as well as blood. 4. The motor nerve conduction velocity and serum folate level were interrelated as a whole including normal and hepatoma rats. 5. The folate administered hepatoma rats were prevented from delaying the motor nerve conduction velocity with normal serum serine/glycine ratio. 6. From the above results, it was conjectured that folate deficiency probably play an important role in developing carcinomatous neuropathy.
