The effects of isoproterenol, PGE1 and NaF on the concentration of cAMP and adenyl cyclase activities in tonsillar lymphocytes, including various stages of tonsillitis, were investigated in comparison with those in peripheral lymphocytes. The increases of cAMP concentration induced by 10-5M isoproterenol and 10-5)M prostaglandin E1 were significantly lower in tonsillar lymphocytes than those in peripheral lymphocytes, although basal cAMP levels were similar in both types of lymphocytes. Tonsillar T lymphocytes had a lower cAMP basal level than B lymphocytes, whereas no difference was observed on the responsiveness to the above agents in either subpopulation. On the other hand, tonsillar lymphocytes from recurrent tonsillitis had significantly lower responsiveness to isoproterenol and PGE1 in respect to cAMP levels than those from other forms of chronic tonsillitis. Furthermore, in tonsillar lymphocytes having had a recent attack of infection, a markedly depressed responsiveness of the cAMP level was recognized. The effects of the above agents on the cAMP level of tonsillar lymphocytes had nothing to do with the age of the patients. Although tonsillar lymphocytes from a hypertrophic tonsil had a low responsiveness to the above agents, other important factors such as various infective states must be further studied in approaching this problem. Tonsillar lymphocytes had a higher basal activity of adenyl cyclase than peripheral lymphocytes, whereas the activations of adenyl cyclase of tonsillar lymphocytes by 10-5M isoproterenol, 10-5M PGE1 and 10mM NaF respectively, were smaller than those of peripheral lymphocytes. From the above results, the changes in the biosynthetic rate of cAMP and the possible role of metabolic modulation of cAMP were discussed in relation to the physiological function of tonsillar lymphocytes which had been exposed frequently to antigens or chemical mediators.
