Changes of mechanical responses of frog single twitch fiber surviving in normal Ringer solution, Ringer solution which was supplemented with antibiotics or modified Medium 199 were examined and the following results were obtained. 1. The peak tensions of twitch and potassium contracture (K-contracture) decreased gradually with the lapse of time after immersion in normal Ringer solution. The time course of inhibition showed two distinct phases; the first phase in which the tension was inhibited slowly and slightly, and the second phase in which the tension was inhibited very rapidly and completely. The twitch was abolished at about 13.3 hours after immersion, and abolishment of K-contracture was observed 30-40 minutes after that of the twitch. However, caffeine contracture did not change significantly at the time when K-contracture was abolished. 2. The time required for the abolishment of twitch and K-contracture of single fiber surviving in Ringer solution with antibiotics was similar to that of single fiber surviving in normal Ringer solution. Caffeine contracture showed no significant changes at the time when K-contracture was abolished. 3. The twitch was completely inhibited at about 6.4 hours after immersion of a single fiber in modified Medium 199. At this time, the peak tension of K-contracture was inhibited by 35% of the control, but caffeine contracture showed no significant changes. 4. When the twitch of a single fiber surviving in normal Ringer solution was abolished and the fiber was then exposed for 20 minutes to modified Medium 199, a recovery of the twitch was not observed. On the basis of these results, the mechanism of inhibition of mechanical responses in surviving single twitch fiber was discussed.
