The effect of endotoxin on lysosomes, lysosomal acid phosphatase and β-glucuronidase was studied in rat river. The initial hepatic changes by a single intraperitoneal injection of endotoxin were seen in Kupffer cells. Within 1 hour after endotoxin treatment, lysosomes in Kupffer cells increased in size and number, and many phagocytic vacuoles containing various materials and organelles were observed. Biochemically, the lysosomal enzymes originating from Kupffer cells began to be activated and released from these cells, and activity of the isozyme of acid phosphatase characteristic of Kupffer cells increased. Such changes in lysosomes were hardly seen in parenchymal cells and endothelial cells at this stage. Parenchymal cells and endothelial cells did not manifest any morphological changes until 2-3 hour later when thrombus formation occurred in the sinusoids. Thereafter, an increase in activity of lysosomal enzymes of originating mainly from parenchymal cells was observed, accompanying by an appearance of autophagic vacuoles in degenerated parenchymal cells. These results indicate that early activation of the lysosomal enzyme in liver was derived from Kupffer cells and the activation of them in parenchymal cells appeared thereafter because of the anoxic state caused by thrombus formation.
