The effects of aldosterone and dexamethasone on blood chemical mediators in endotoxin shock were・studied. The results were as follows. 1) Plasma histamine concentration after lethal dose endotoxin administration was elevated with 1-5min. The peak of plasma histamine cocentration was in 15min. Plasma histamine concentration was elevated again in 60min. 2) Plasma serotonin concentration after lethal dose endotoxin administration was elevated within 1min and again in 5min. Plasma serotonin concentration was still elevated between 5 and 180min. 3) Blood bradykinin concentration after lethal dose endotoxin administration was elevated within 1min and it reached the maximum. Blood bradykinin concentration was diminished between 4 and 30min and then elevated after 60min. 4) Plasma adrenaline concentration after lethal dose endotoxin administration was slowly elevated within 5min and maintained the same level for 60min after which it elevated again up to 180min. 5) Plasma noradrenaline concentration after lethal close endotoxin administration was slowly elevated within 5min and maintained same level for 30min. Plasma noradrenaline concentration was elevated again in 60min. 6) Mortality rate was determined 48 hrs after lethal dose endotoxin. Aldosterone resulted in a mortality rate of 80per cent. Dexamethasone resulted in a mortality rate of 55.2per cent. Aldosterone plus dexamethasone resulted in a mortality rate of 17.9per cent. 7) Injection of endotoxin was followed by an immediate, profound hypotension for 1 to 5min, a secondary rise in pressure and then a subsequent fall in pressure 60min later. All rabbits died, usually within 5 to 9 hrs. After aldosterone and dexamethasone administration, the blood pressure of the rabbits fell gradually. 8) Dexamethasone blocked the elevation of plasma histamine, plasma serotonin, blood bradykinin and plasma catecholamine concentration in endotoxin shock. 9) Aldosterone slightly blocked the elevation of plasma histamine, plasma serotonin, blood bradykinin and plasma catecholamine concentration in endotoxin shock. 10) Dexamethasone and aldosterone (mixed) completely blocked the elevation of plasma histamine, plasma serotonin, blood bradykinin and plasma catecholamine concentration in endotoxin shock. Based on these results, simultaneous administration of dexamethasone and aldosterone may be considerd to inhibit the secretion of chemical mediators and then prevent endotoxin shock.
