In order to support an activity of conservation of valuable natural population and biological diversity in aquatic environment, the valuable population genetic data are necessary. In our laboratory, Department of Aquaculture-Kochi University, beside allozymes which have been established for more twenty years ago for revealing those genetic data, some molecular genetic techniques such as Random Amplified Polymorphic DNA (RAPD); Multilocus DNA Fingerprinting; Amplified Restriction Fragment Polymorphism (AFLP); and Variable Number Tandem Repeats (VNTR) of Microsatellite, have also been used.During study of the DNA polymorphism in several fish species, we conclude that the RAPD and multi-locus DNA fingerprinting (including AFLP) analysis are most useful for pedigree study or species identification, while population genetic parameters will be more accurately estimated by single locus of microsatellite DNA. This paper shows a protocol for detection of fish DNA polymorphism in our laboratory.
