Quenching of pyrene and phenothiazine fluorescence by methylviologen was studied in dihexadecylphosphate vesicle solutions. Methylviologen in the outer aqueous phase of the vesicle quenched the fluorescence more strongly than that in the inner aqueous phase. The Stern-Volmer quenching constants of pyrene fluorescence were determined to be 7.3 x 10^4 and 1.1 x 10^4 M^<-1> for the solutions containing methylviologen in the outer and inner aqueous phase, respectively. The difference in the quenching constants was then utilized to estimate the transmembrane transport of methylviologen from the inner aqueous phase to the outer one. No transmembrane transport occurred while the solution was kept at the room temperature for a week. And further, the leakage of methylviologen through the vesicle bilayer was not observed even when the solution was submitted to the freezing-thawing-sonication treatment.
