Spleen cells from a Balb/c mouse immunized with a human myeloma line ARH-77 were fused with cells of the mouse myeloma line NS-1. A monoclonal antibody HMy-1 continuously secreted by established hybridoma cells recognized a new cell surface antigen specincity. This newly found antigen was selectively expressed on cells in myeloid lineage, but not on lymphocytes or monocytes, when tested against cells from lymph node, tonsil, thymus, bone marrow and peripheral blood cells including erythrocytes and platelets. Similarly, on the studies tested against cells of 98 patients with leukemia and lymphoma, the HMy-1 antibody reacted with those from patients with myeloid leukemia, but not with those from patients with acute lymphocytic leukemia or lymphoma. An unexpected exception to this pattern was the weak reaction of HMy-1 antibody with cells from patients with a B-cell type chronic lymphocytic leukemia. The studies on myeloid leukemia allow for the following conclusion. 1) Almost all cases of chronic myelocytic leukemia (CML) were positive for the HMy-1 antigen. 2) Almost allcases of CML in myeloid crisis were also positive for the HMy-1 antigen. 3) Around half of the cases of acute myeloid leukemia (AML), which were mainly characterized as the M1 and M2 type AML in the FAB classification, preferentially expressed the HMy-1 antigen. The inhibition studies of the monoclonal antibody on colony formlng assay suggested that the HMy-1 might be expressed on both grnulocyte-macrophage colony-forming cells (CFU-GM) and erythroid colony-forming cells (CFUE). Immune precipitates formed with HMy-1 antibody and detergent lysates of radiolabelled CML cells contained a single polypeptide with a molecular weight of 99000 daltons. In consideration of these results, the HMy-1 antibody may provide a useful tool for the diagnosis of acute leukemia, especially of CML in myeloid crisis and moreover for the study of the heterogeneity and myelocyte-monocyte differentiation, which might be underlying in AML.
