Passenger Leukocytes (PL) derived from grafted organs play an important role in initiation of immune responses after transplantation. In the earliest stage of transplantation, PL expressing allo-antigen migrate into recipient tissues from the grafted organ. Then PL are recognized by allo-reactive T cells of the recipient and immune response is induced. Skin grafts are rejected even in the presence of strong immuno-supressive protocols; however acceptance of heart grafts is more easily induced in many cases. It was suspected that PL might play an important part in the reaction in host organs following a graft (organ specificity). We constructed mouse skin and heterotopic heart transplantation models using female C3H/He mice as recipients and male C3H/He mice as donors. Donor PL were measured by the polymerase chain reaction method using male specific primer encoded Zfy-1 gene at days 2, 5, and 10 after transplantation. Donor derived PL following heart transplantation (HPL) were detected in all analyzed tissues: thymus, spleen, liver, mesenteric lymph node, peripheral blood, skin, and bone marrow (38.5% of analyzed samples). On the other and, hand donor derived PL in skin transplantation (SPL) were detected only rarely and at very low levels in blood, spleen, and skin (6.0% of analyzed samples). These results demonstrated that SPL were distributed only around the transplanted skin, while HPL migrated throughout the body. It seems probable that different characteristics of migration by PL establish the organ specificity that determines graft acceptance or rejection in transplantation.
