The levels of surfactant protein (SP)-A, SP-B and SP-D were studied in bron-choalveolar lavage (BAL)-subfractions and lamellar body (LB) subfractions of rats with chronic lung injury induced by intratracheal exposure of silica (40 mg/kg of body weight) to rats. Silica- or saline-treated rats were sacrificed on day 7, day 14 and day 28, and BAL and LB were obtained. The BAL was divided into two subfractions by two different methods, i. e, alv1 and alv 2 fractions as defined by Nicholas et al, and LA (large aggregates) and SA (small aggregates) fractions accord-ing to Jobe et al. The LB was divided into two subfractions, lbA and lbB fractions according to Nicholas et al. These BAL-subfractions and LB subfractions were analyzed for total protein, total phospholipid and surfactant proteins such as SP-A, SP-B and SP-D. The results obtained are sum-marised as follows : 1) The levels of total protein and phospholipid in alv 2 and SA fractions increased in silica-treated rats through day 14, indicating that pool sizes of small aggregate-formed surfactant (SA-Sf), which represented nearly the totality of the phospholipid content, had increased in the alveolar space, but until day 28, they remained unchanged in the alveolar space in the chronic lung injury. This result suggests that at first conversion of large aggregate-formed surfactant (LA-Sf) to SA-Sf might have been induced in the alveoli, but finally the LA-Sf accumulation rate might exceed the rate of conversion of LA-Sf to SA-Sf in the chronic lung injury. 2) In silica-treated rats, SP-A and SP-B levels in LA-Sf and lbB subfractions increased mark-edly during the experimental period, indicating that Sf is synthesized markedly by alveolar type II cells and accumulates in the alveolar space of silica-treated rats. 3) Changes of SP-A, SP-B and SP-D levels in BAL-subfractions of silica-treated rats varied from each other. SP-A increased markedly in LA fraction, while the increase of SP-B in LA frac-tion was relatively small. This result suggests that SP-B is also degraded in the alveolar space during the chronic lung injury. SP-D increased markedly in SA fraction of silica-treated rats.
