In order to analyze mechanisms controlling infiltration of T lymphocytes into tumor tissues, HEV cells were isolated from cervical lymph nodes of Fischer rats and incubated with lymphocytes under stimulation by various cytokines including lymphocyte migration factor-b (LMF-b). LMF-b augmented the binding of lymphocytes to HEV cells. Furthermore, LMF-b was not only chemotactic for CD8+ T cells, but augmented the binding of CD8+ T cells to HEV cells. The augmented binding by LMF-b was inhibited by the anti-lymphocyte function associated antigen-1 (LFA-1) antibody, anti-CD2 antibody, and fibronectin derived peptide, CS-1. It was suggested that the augmentation of lymphocyte adhesion to HEV cells by LMF-b was mediated by the lymphocyte function associated antigen-1/intercellular adhesion molecule-1 (LFA-1/ICAM-1) pathway, the CD2/LFA-3 pathway, and the very late antigen-4/CS-1 (VLA-4/CS-1) pathway.
