Parietal cell antibodies (PCA) are often found in sera of patients with autoimmune gastritis (type A gastritis) and pernicious anemia. In this study, we produced the recombinant gas- tric H?, K?-ATPase protein to define the target molecule of PCA. 1. We obtained the cDNA fragment of gastric H?, K?-ATPase by reverse transcriptase polymer- ase chain reaction (RT-PCR) from the total RNA of gastric biopsy samples. This cDNA fragment encodes residues 497-743 of the α subunit of gastric H?, K?-ATPase, which are thought to be part of its catalytic site. A 52kD recombinant fusion protein of the α subunit of gastric H?, K?-ATPase was produced in E. coli using this cDNA fragment. 2. Thirty human sera containing PCA were studied by immunoblot analysis. Twenty (66.7%) reacted with the 52 kD recombinant protein. 3. The intensity of immunohistochemical staining of parietal cells in rat gastric mucosa by PCA positive serum was reduced by absorption of serum PCA by the 52 kD recombinant protein. 4. Three of 4 sera of patients with type A gasritis reacted with the 52kD recombinant protein, but the sera of 11 patients with type B gastritis did not react with the 52kD recombinant protein at all. These results suggest that a part of PCA recognizes the α subunit of gastric H?, K?-ATPase, that the epitope exists in the region containing its catalytic site, and that PCA might be involved in the pathogenesis of autoimmune gastritis (type A gastritis).
