Serum IgM antibody, which is immunohistologically reactive with the basal aspect of syncytiotrophoblast (BAST)of normal human placenta, was investigated by immunohistological and immunoblotting methods. The corresponding antigen of anti-BAST antiserum was also examined in leukemia cells, breast cancer tissues and circulating immune complex (CIC). Results were as follows. 1) The anti-BAST antibody was often detected in the sera of patients with acute leukemia and breast cancer. Its titer and positivity was significantly higher in patients at the early stage than at the late stage. 2) Electron microscopic observation of leukemic cells from an anti-BAST positive patient after treatment of the cells by retrovirus-activator BrdU demonstrated the occurrence of retrovirus-like particles. 3) A retrovirus-related antigen was detected in circulating immune complex obtained from anti BAST positive sera by Western blot analysis. 4) Anti-BAST positive sera from patients with acute leukemia reacted with cryostat sections of breast cancer tissues. Immunofluorescence absorption analysis suggested that the corresponding antigens recognized by the serum antibodies might be the same as retrovirus-related antigens which are present in the BAST. 5) Anti-BAST antibody was frequently detected in the sera of the victims of the atomic bomb at Hiroshima in 1945. Follow-up studies for 5 years revealed significantly higher occurence of malig- nancies in the anti-BAST positive subjects than in the negative ones.
