Methylcholanthrene誘導A線維肉腫細胞由来低転移クローンへのfyn導入による実験的肺転移能増強の試み

Accumulating studies of metastasis concerning oncogenes have been provided by recent development of the molecular biological approach. However, few reports comparing on- cogene expression between high-and low-metastatic clones have been raised. In the present study, the expression of seven different oncogenes (ras, myc, erbB2, src, fyn, yes and sis) were examined for a high-metastatic clone (MH-02) and its parent clone (ML-01) derived from murine methylcholanthrene induced A flbrosarcoma (Meth A). The result showed that fyn oncogene and sis oncogene were expressed at higher levels in MH-02 than in ML-01 cells, being 3.6 times higher for the former and 1.8 times higher for the latter. On the basis of this result, expression vector containing fyn cDNA was transfected into ML-01 cells by electroporation and the transfectants were administrated to mice through the tail vein to evaluate their metastatic potential by counting lung metastatic nodules. The result indicated that a 1.5-2 fold increase in metastatic potential was noticed in the fyn-expressing transfectants as compared to ML-01 cells transfected with control vector with no fyn gene. The result disclosed that fyn oncogene is one of the factors associated with metastatic potential of Meth A.
