Changes in the phenotypic and transcriptional properties of HuH-6, a human he- patoblastoma cell line, were investigated by treatment with dibutyryl cyclic AMP (dbcAMP), treat- ment with retinoic acid (RA), or co-culture with flbroblastic cells. Treatment of HuH-6 cells with either dbcAMP or RA inhibited the expression of alpha-fetoprotein (AFP) mRNA, while it en- hanced the expression of albumin mRNA. Such treatment also induced the expression of connexin 32 (cx32) mRNA. Immunohistochemical examinations of the HuH-6 cells with an anti-cx32 antibody showed a marked increase in the number of gap junctions after treatment with these agents as well as after co- culture with BALB/c 3T3 cells. Dye-transfer assay, using fluorescent Lucifer Yellow microinjec- tion, revealed that induced cx32 proteins function as gap junctions. Immunohistochemical examinations also demonstrated that dbcAMP or RA treatment and co- culture with BALB/c 3T3 induced increased expression of vimentin and localization of desmoplakin at the plasma membrane. These findings strongly suggested that treatment of HuH-6 cells with dbcAMP or RA, and co- culture with BALB 3T3/c changed the phenotypes and other properties of the hepatoblastoma cell toward differentiation.
