Using CA 125 producing human ovarian cancer cell lines, the ratio of CA 125 positive cells was examined by immunocytochemical method. Further, cellular proliferation, the secretion of CA 125 and change in its production were studied following the addition of various drugs. Secretion of CA 125 decreased at the logarithmic growth phase and increased during both the lag phase and transition to the stationary phase. DBc-AMP, theo, NaBT and TPA were confirmed to increase the production and secretion of CA 125, when compared with corti, which had opposite effect. DBc-AMP and theo had similar effects on the time-course of the production and secretion of CA 125, suggesting the involvement of A kinase. Administration of NaBT led to a strong inhibition of cellular proliferation and to such an increase in the secretion of CA 125 that almost all cells were conflirmed to be CA 125 positive cells. This suggests that the action of NaBT changed the CA 125 negative cells to positive ones. Combined use of sphingosine and TPA led to only slightly lower secretion of CA 125 than with TPA alone, suggesting slighly less involvement of C kinase. With TPA alone there was no change in ratio of CA 125 positive cells, but an enhancement in the degree of positiveness was detected. Corti was confirmed to inhibit temporarily the production and secretion of CA 125 and to decrease the positiveness of the CA 125 positive cells. The production of CA 125 thus appeared to be modified by a number of drugs having different mechanisms of action.
