This study was undertaken to determine the effects of caerulein (CA) and hydrocortisone (HC) administration by single use, combined use, or separate use on the regeneration of the remnant pancreas after pancreatectomy in rats. The results obtained were as follows: 1) In single administration, the improvement of endocrine function was observed in the 90% pancreatectomy group with the administration of either CA 3.0 μg/kg/day or HC 2.0 mg/kg/day, but a more diabetic state appeared in the 90% pancreatectomy group with administration of HC 5.0 mg/kg/day. The regeneration of exocrine cells was observed in both the 70% and 90% pancreatectomy groups with administration of CA 3.0 μg/kg/day, HC 2.0 mg/kg/day or HC 5.0 mg/kg/day. 2) In combined administration of CA 0.3 μg/kg/day and HC 2.0 mg/kg/day, the improvement of endocrine function was observed in the 90% pancreatectomy group at the 28 days after operation, but the result was similar to that of HC 2.0 mg/kg/day single administration. The effect of the combined administration of CA 3.0 μg/kg/day and HC 2.0 mg/kg/day on the improvement of endocrine function was better than single administration at the 28 days after operation, and the improvement was also observed at the 56 days after operation. However, it was not enough to prevent the appearance of Sandmeyer's diabetes. The regeneration of exocrine cells at the 56 days after operation became better in combined administration than in single administration. 3) In separate administration of CA 3.0 μg/kg/day and HC 2.0 mg/kg/day, the improvement of endocrine function and the regeneration of exocrine cells were observed in the 90% pancreatectomy group at the 28 and 56 days after operation. Separate administration was more effect than single administration, especially with HC-former administration, where HC was given prior to CA. These data suggested that the combined or the separate administration of HC and CA on the regeneration of the remnant pancreas after pancreatectomy was more effective than that of single use.
