This study was undertaken to investigate the proliferation and the functional changes of endocrine and exocrine pancreas during the regeneration of the remnant pancreas after 70% or 90% pancreatectomy in rats. The results obtained were as follows: 1) The increase in DNA synthesis was observed in all kinds of cells during the 3~10 days after pancreatectomy. And DNA synthesis was more in 90% pancreatectomy than in 70% pancreatectomy. This phenomena of exocrine cells slightly preceded that of islet cells. 2) The biochemical data on tissue amylase activity, protein and DNA contents suggested the growth of exocrine cells during the 28 days after operations. This tendency was observed in both pancreatectomies. 3) In the pancreatic function diagnostant test as an exocrine function test, normalization in the 70% pancreatectomy group was observed 21 days after operation, but not in the 90% group during the observation time. 4) The results in the quantitation of islet hormone contents in the parabiliary segment suggested the growth of endocrine cells during the 28 days after operation. However, a difference in regenerative reaction was observed among each constituent cell of islets. Namely, the regeneration of A cells was more than that of B or D cells. This tendency was shown in both pancreatectomies, but in 90% pancreatectomy, the content of hormone produced by A, B and D cells began to decrease from 28 days after operation. 5) Diabetic condition was observed temporarily at the early stage in the 70% pancreatectomy group, while that in the 90% pancreatectomy group was observed throughout the entire course after operation. In 90% pancreatectomy, there was a gradual improvement of the diabetes during the first 28 days after operation, but Sandmeyer's type diabetes was induced thereafter. Thus, it was concluded that some differences exist between 70% pancretectomy and 90% pancreatectomy, and between endocrine and exocrine pancreas in regeneration after pancreatectomy. The parallel growth of endocrine cells and exocrine cells after pancreatectomies was confirmed.
