We have already established and reported a pair of human autologous tumor specific cytotoxic T lymphocyte (CTL) clone TCHMC-1 and target cell clone HMC-1-8, both of which were derived from the metastatic pleural effusion of a female patient with breast cancer. In this paper, we describe the target antigen expressed on HMC-1-8 that was defined by monoclonal antibody (mAb) 3A2. This mAb inhibited the cytotoxicity of TCHMC-1 against HMC-1-8 cells selectively, but did not inhibit the cytotoxic action of LAK cells against HMC-1-8 cells. Western blot analysis using mAb 3A2 identified a molecule with approximately 92 kDa M. W. The 3A2-defined antigen was highly expressed on autologous primary cancer tissue, but not on the normal mammary gland tissue in the same patient. Moreover, this antigen can be detected on more than 50% of the human allogeneic breast cancers, but not on other neoplastic cells except for one out of seventeen prostatic cancers. Non-neoplastic normal cells seldom expressed this antigen. It was also suggested that 3A2- defined antigen is not a murine mammary tumor virus-related product. These data suggest that this antigen is a candidate for a tumor specific antigen which could be the target molecule of human autologous CTLs.
