Expression of ras p21 in human uterine cervical cancer, benign and precancerous lesions (dysplasia) and normal squamous epithelium was immunohistochemically evaluated by the avidin-biotin complex (ABC) immunoperoxidase method with anti-ras p21 mouse monoclonal antibody rp-35. The positive p21 rate was 0% in normal squamous epithelium, 10.0% in squamous metaplasia, 17.9% in mild dysplasia, 28.9% in moderate dysplasia, 47.8% in severe dysplasia, 56.6% in carcinoma in situ, and 50.0% in microinvasive carcinoma. In the follow-up studies of patients with mild or moderate dysplasia, ten regressive cases showed 20.0% ras p21 positivity, but 14 progressive cases which developed into more malignant lesions showed 50.0% ras p21 positivity. Furthermore, we studied the reactivity of 170 squamous cell carcinomas of the uterine cervix of various histological types and 11 regional metastatic lymphnodes. The expression of p21 was noted in 57.1% of keratinizing type, 54.2% of large cell non-keratinizing type. 38.7% of small cell non-keratinizing type, and 54.5% of regional metastatic lymphnode. There was no significant difference between the p21 positivity and clinical stage or lymphnode metastasis. It was suggested that the intervention of ras oncogene in human uterine cervical cancer occurred in early carcinogenesis more profoundly than in progression of cancer and in metastasis of regional lymphnode.
