Tissues from cervical carcinomas and precancerous lesions were examined for the presence of human papillomavirus DNA, and the physical state of HPV16 DNA in cervical carcinomas was analysed. 1) HPV DNA was detected in 62% (28/45) of cervical carcinomas in the Hokkaido area. HPV16 DNA was found in 54% (24/45), HPV18 DNA in 18% (8/45) and HPV33 DNA in 7% (1/14), whereas none of 11 cases of cervical carcinomas contained HPV6/11 DNAs. In cervical dysplasias and carcinomas in situ, HPV16 DNA was detected in 40% (12/27), HPV18 DNA in 7% (2/27) and total HPV DNAs in 48% (13/27). HPV6/11 DNA was not present in 5 cases. 2) In HPV16 DNA positive cases of carcinoma, we analysed the patterns of digested fragments, after digesting the total cellular DNA with restriction enzymes (BamHI+PstI). Three cases were found to have lost the fragment A or both the fragment A and D with appearance of off-sized fragments instead. In contrast the fragment B, for example, was commonly detected as a band of same size with the corresponding HPV16 DNA fragment in the marker lane. In these cases HPV16 DNA may be integrated in the host cell DNAs. 3) The physical state of HPV16 DNA was examined with two-dimensional gel electrophoresis in 10 cases of cervical cancers containing HPV16 DNA. In nine cases HPV16 DNA was integrated in the host cell DNA, but one case contained only episomal HPV16 DNA sequences. These results suggest that the integration of HPV16 DNA mainly play an important role, but the episomal HPV16 DNA can also play some role in carcinogenesis.
