Employing in vitro cultured and virus-infected autologous monocytes as target cells, responses of cytotoxic lymphocytes (CTL) to respiratory syncytial virus (RSV) infection were studied after peripheral blood lymphocytes were cultured with the virus antigen. In control groups, newborn infants exhibited little CTL activity while a portion of normal children aged one or over and adults who were examined during the RSV-epidemic season had CTL activity exceeding 10% with specific lysis. Twenty-five of 40 children with RSV infection developed CTL activity during the observation period. The CTL responses, however, were shown to be age-dependent since in the initial few weeks the average activity (10.8±17.0%) in infants less than 6 months old was significantly lower than that (32.7±31.3%) of the older subjects (p<0.01). The rise of CTL activity generally coincided with the cessation of RSV shedding and a few subjects with severe illness were shown to have rather low CTL activity. Reactivity of CTL to allogeneic target cells suggested that the major CTL activity may be due to the cytotoxic T cells. These observations seem to indicate the close association of CTL activity with recovery from RSV infection.
