On the basis of an earlier observation that tumor necrosis factor (TNF) activity in the serum of nude mice is extremely low, the role of the priming agent (P. acnes) in the production of TNF in relation to the function of T cells was investigated in Balb/c, nu/nu mice and those inoculated with thymus (T cell reconstituted nude mice). The role of eliciting agent (LPS) in TNF production was also investigated employing the dot blot analysis for expression of TNF-mRNA in spleen macrophage from Balb/c, nu/+ mouse. The results obtained were as follows: 1) TNF activity in serum from Balb/c, nu/nu mice was approximately 1/22 compared to that from Balb/c, nu/+ mice. 2) TNF activity in serum of T cell reconstituted nude mice as compared to that of Balb/c, nu/nu mice showed a 14 fold increment. 3) TNF activity in conditioned medium of the splenic macrophage from Balb/c, nu/+ was substantially high while that of T cells was not detectable. The conditioned medium of splenic macrophages from Balb/c, nu/nu exhibited TNF activity almost as high as that from Balb/c, nu/+. 4) By administration of P. Acnes, the population of splenic macrophages of Balb/c, nu/+ mice increased 25.5 times, whereas that of Balb/c, nu/nu mice increased 6.8 times. 5) The splenic macrophages of Balb/c, nu/+ mouse administered with P. acnes expressed approximately 3-4 folds more TNF-mRNA by treatment with LPS in vitro than those which were LPS non-treated. The above results suggest that T cells are involved in the proliferation of splenic macrophages upon the administration of priming agent (P. acnes) and that the eliciting agent stimulates the expression of TNF-mRNA in the macrophages.
