The adhesive properties of MH-02 clone to type IV collagen and endothelial cells were investigated in comparison with that of a low metastatic clone (ML-01). The number of MH-02 cells attached to either type IV collagen or calf pulmonary endothelial cell was significantly higher than that of ML-01 cells. The difference of adhesiveness between MH-02 and ML-01 cells was apparently obliterated by preincubation of these cells with antibody F(ab') 2 against MH-02 which had been preabsorbed with ML-01. In an attempt to clarify if the adhesive property of the MH-02 cell observed in vitro experiments was related to its enhanced metastatic potential, tumor cells were first incubated with the anti-MH-02 F(ab') 2 and injected into BALB/c mice via tail vein. Not only number of metastatic nodules in the lung but also the rate of tumor arrest measured by incorporation of 125I-IUDR into the lung was substantially reduced by treatment with the antibody. The reduction of metastatic nodules or tumor arrest was not likely due to the tumoricidal effect of anti-MH-02 F(ab') 2 since neither cytotoxicity nor inhibition of tumor growth was noticed by in vitro experiment. The radiolabeled membrane components of MH-02 cells which reacted with this antibody were then analysed by SDS-PAGE and autoradiography and several bands (mainly of 95Kd and 180Kd) were identified. Physicochemical characterization of these membrane components is currently under investigation.
