γ-Glutamyl transpeptidase (GGT) was solubilized from squamous cell carcinoma tissues of human skin by Triton x-100 and bromelain treatment, and some of its biochemical properties were compared with those of normal skin and normal kidney. Activities of the enzyme from squamous cell carcinoma tissues were 20-100 times higher than those of normal skin enzyme. However the enzyme from the skin, squamous cell carcinoma and kidney were found to be very similar with the optimal points of pH, Km value and effects of various L-amino acid acceptors. The enzyme from squamous cell carcinoma was distinguished from normal tissues by increased affinity to concanavalin A, iso-electric focusing patterns and mobility on gradient polyacrylamide gel electrophoresis. These results suggest that the isozymic form of squamous cell carcinoma enzyme is mainly due to the altered glycosylation of this glycoprotein.
