A novel cell-surface antigen (MET-1) on rat lymkhto-hemopoietic cells was determined by using a monclonal antibody, R2-1B3 (1B3). The 1B3 antibody was tested for its reactivity with different hemopoietic cells by cytofluorography with a FACS analyzer. It labeled more than 80% of the lymph node, spleen and bone marrow cells, and 10-20% of the thymus cells. Cytofluorographic analysis performed on purified rat T cells, B cells, macrophages, and granulocytes demonstrated that MET-1 antigen was readily detectable on all of these cell types, with the greatest expression on B cells. A minor population of thymocytes associated with MET-1 antigen appeared to be cortisone-resistant and were located mainly in the thymic medulla. These MET-1 (+) thymocytes seemed to be functionally more mature than MET-1 (-) thymus cells, as suggested by the fact that the cytotoxic treatment of thymus cells with 1B3 antibody and complement (C) resulted in a significant reduction of their proliferative responsiveness to phytomitogens and lymphokines (CAS) derived from concanavalin A (con A) activated rat spleen cell culture. Immunochemical data showed that 1B3 antibody recongnized the ill defined band with a molecular weight of 27K to 42K daltons estimated by SDS-polyacrylamide gel electrophoresis. In the following experiment, we have attempted to induce MET-1 antigen on MET-1 (-) thymocyte population. For this porpose, rat thymocytes depleted of MET-1 (+) thymocytes by repeated cytotoxic treatment with 1B3 and C were cultured in vitro in the presence of either CAS or con A plus interleukin 2 (IL-2). MET-1 antigen could be induced on these MET-1 (-) thymocytes, in correspondence with their activation and expression of IL-2 receptors defined by ART-18 antibody. When MET-1 (-) thymocytes were separated into peanut agglutinin (PNA)-positive and PNA-negative populations and tested for MET-1 induction in each fraction with CAS or con A plus IL-2, it was found that MET-1 could be induced in the PNA(-) but not in the PNA(+) population. Immuno-biological significance of differentiation from MET-1 (-) PNA(-) thymocytes into MET-1 (+) PNA(-) thymus cells was discussed in relation to the intrathymic differentiation pathway of rat T cells.
