Chondrocytes from articular cartilages of young rabbits were isolated using collagenase treatment. The chondrocytes retained their characteristics in vitro which differentiated them from fibroblasts, even after 1-week of culture. The lipid composition of the fresh and 1-week cultured cells was analyzed. The results obtained are summarized as follows; Main lipids of the fresh cells were phospholipids and triacylglycerol(TG). The phospholipid content was 2.8 fold of TG. The most prominent phospholipid was phosphatidylcholine(PC), the great majority of which was of a diacyl type. Other phospholipids occurred in the order of phosphatidylethanolamine(PE)>phosphatidylinositol(PI)=phosphatidylserine(PS)>sphingomyelin(SM), respectively. The phospholipids contained oleate as the main fatty acid, and the contents of linoleate and arachidonate were relatively low. PC was mainly composed of molecular species containing oleate, i. e., 16:0/18:1, 18:0/18:1, 18:1/18:2 and 18:1/18:1 species. Molecular species containing arachidonate were less, that is, 16:0/20:4 was 3.1% and 18:0/20:4was 6.2%, respectively. The characteristics of lipid profiles in the fresh chondrocytes were not changed qualitatively and quantitatively in the cells cultured for 1-week at a low concentration of essential fatty acids. These lipid profiles of articular chondrocytes, which are high oleate, as well as low linoleate and arachidonate, seem to reflect specific routes of nutrient uptake of the chondrocytes. Articular chondrocytes might bring a limited supply of essential fatty acids.
