It has been reported that IFN enhances IgE-mediated histamine release from basophils. Histamine is one of the chemical mediators to cause an asthma attack. Histamine plays an important role, especially in atopic asthma. Therefore, the relationship between atopic bronchial asthma which belongs to the Coombs and Gell's type 1 allergy, and the IFN-system was investigated. IFN was measured by semi-micro dye-binding assay. Oligo-2', 5'-adenylate synthetase (2', 5'-AS) in peripheral lymphocytes (PBLs) which is known as one of the IFN-induced enzymes was measured by the liquid solution method. Serum IFN was detected in 40.6% of asthmatic patients, but not in most of the healthy control. It was suggested that most of the serum IFN detected in asthmatic patients is α-IFN and the rest may be γ-IFN by pH 2.0 dialysis and anti-α-IFN treatment. Furthermore, activity of 2', 5'-AS in asthmatic patients was higher than in healthy control (p< 0.05). These results of serum IFN and 2', 5'-AS suggest that IFN productive capacity in asthmatic patients may be increased. Therefore, IFN productive capacity was investigated with PHA, ConA, OK432 and HSV. IFN produced in PBLs of asthmatic patients was significantly higher than in those of healthy control in the cases of PHA, ConA and HSV (p<0.05). In the case of OK432, there was no significant differe-nce between asthmatic patients and healthy control, but IFN produced in PBLs of asthmatic patients tended to be higher than in healthy control (0.05<p<0.1). These results show that α- and γ-IFN productive capacities in asthmatic patients are increased. Therefore, we investigated the influence of α- and γ-IFN on histamine release from basophils. IFN was added to peripheral leukocytes and the cells were incubated for 6 hours. The cells were then treated with anti-IgE. The released histamine was measured by fluorometric assay. As a result, α- and γ-IFN enhanced the IgE-mediated histamine release. The function of γ-IFN was significantly stronger than α-IFN (p<0.05). These results show that the IFN-system may be closely related to bronchial asthma.
