The segments of leaves from log sprouts of Keyaki (Zelkova serrata) were cultured on modified MS medium supplemented with BAP (1, 2 and 5mg/l). Each medium induced adventitious buds. They were induced on callused tissue or directly on the surface of the explant. Adventitious bud formation rate was highest on the medium containning 2mg/l of BAP. After 4 weeks of culture, explants became brown and died. A high concentration of BAP decreased the browning and death of explants. Explants which induced adventitious buds were transferred to modified MS medium with 1mg/l of BAP. Shoot elongation occurred on some explants. Shoots were rooted on half strength of modified MS medium supplement with IAA (1mg/l) or IBA (0.1, 0.5mg/l).
