[6]-Gingerol, the pungent principle component of ginger, has been reported to have pharmacological activity such as antioxidative activity, inhibitory activity against a prostaglangin biosynthesizing enzyme, antiinflammatory effect, antifebrile effect, and analgesic effect. We synthesized gingerol analogues containing novel compounds via terminal-esteric shogaol as a key intermediate. Furthermore, we examined the inhibitory activity of gingerol analogues toward tyrosinase that is known to be a key enzyme for melanin biosynthesis. The activity of the examined gingerol analogues were shown to be dependent on side-chain structures. More effective compounds contained a w-carboxyl group, and one hydroxyl or carbonyl group on the internal site of side-chain. Also it seems that a 1,3-diol structure on the side-chain was effective but a β-hydroxy ketone structure was negative to inhibitory activity toward mushroom tyrosinase.
