The role of adenosine A_1 receptor (A_1R) in cardioprotective effects against ischemia has received increased attention. In the present study, in order to investigate whether ischemia or ischemic preconditioning affects gene expression of A_1R in the heart, we first cloned heart A_1R from rat and human using the polymerase chain reaction (PCR) method and examined the expression of A_1R mRNA in rat hearts with/without ischemic preconditioning by Northern blot probed with the rat heart A_1R cDNA. The cDNAs obtained from both species encoded proteins of 981 nucleotides, which were 100% identical to the previously cloned rat and human brain A_1Rs, respectively. The human A_1R showed 89% sequence identity to the rat A_1R at nucleotide level. Expression of heart A_1R mRNAs in Xenopus laevis oocytes showed the functional activities coupled to the inhibition of adenylyl cyclase, which are characteristic to native A_1Rs. Northern blot in the rat showed that the expression of A_1R mRNA was higher in the brain than in the heart. Though it also revealed that its expression in any case were higher than that in control non-ischemic heart, the expression in the heart with preconditioning was obviously higher than that without preconditioning. These results indicate that the transcriptional upregulation of A_1R may be implicated in the cardioprotective effect of ischemic preconditioning.
