The ATP-sensitive potassium (K_<ATP>) channel may contribute to determination of the membrane potential in smooth muscle cells of the ductus arteriosus (DA) and, thus, to regulation of contractile tone in response to oxygen. Oxygen-sensitive, voltage-gated potassium channels (Kv) may also be important in the regulation of ductal tone. This study aimed to investigate developmental changes in the expression of K_<ATP> and Kv in the fetal rat DA. The DA was isolated from fetal rats at days 19 and 21 of gestation (term ; 21.5 days). The expression of K_<ATP> and Kv 1.2, 1.5, 2.1 and 3.1, putative oxygen-sensitive Kv channels, were evaluated at the mRNA and protein levels, using quantitative real-time PCR and immunohistochemistry. Expression of mRNA and protein of K_<ATP> in the 21-day fetus was greater than in the 19-day fetus. Expression of mRNA and protein of Kv in the 21-day fetus was less than in the 19-day fetus. The Kv current in the 21-day fetus was significantly greater than that in the 19-day fetus at 60 mV. These data support the hypothesis that K_<ATP> is important in the regulation of ductal constriction in response to O_2.
