The serum alpha-fetoprotein (AFP), human chorionic somatomammotropin (HCS) and amniotic fluid AFP heve been setimated by radioimmunoassay in normal and complicated pregnant women. The initial appearance of demonstrable serum AFP was noted at 10 weeks of gestation. Subsequently, the serum AFP increased to reach its maximum at approximately 29 to 34 garadually increased with the progress of pregnancy. The amniotic fluid AFP, on the other hand, was at its maximum between 13 and 15 weeks of gestation, and then repidly decreased with the progress of pregnancy to term. Although a slight correlation was found between the serum AFP and HCS at term, the serum AFP and amniotic fluid AFP were in no particular way correlated. The maternal serum AFP may be influenced not only by the fetus itself but also by the placental function during pregnancy. Accordingly, if the AFP is abnormally increased or decreased for the gestational week, it is suggestive of some abnormality in feto-placental relationship. The amniotic fluid AFP runs parallel with the fetal serum AFP, the latter directly reflecting the fetal well-being, Hence, the determination of the amniotic fluid AFP may be considered of value.
