To investigate the role of prostaglandins (PGs) in human labor and ripening of the uterine cervix, the concentrations of prostaglandin E_1 (PGE_1) and prostaglandin F_<2α> (PGF_<2α>) in human myometrial and cervical tissues during pregnancy were measured by radioimmunoassay. In addition, the concentrations of PGE_1 and PGF_<2α> in myometrial and cervical tissues of non-pregnant women treated with various steroid hormones were also determined in order to study the effects of these hormones on the production of PGs. In non-pregnant women, there was no significant difference between the concentrations of PGE_1 and PGF_<2α> in myometrial and cevical trissues. In the first and second trimesters of pregnancy, the concentrations of PGE_1 and PGF_<2α> in myometrial and cervical tissues indicated slightly high levels as compared with those in non- pregnant women. In the third trimester of pregnancy, the concentrations of PGE_1 and PGF_<2α> in cervical tissues represented a significant increase (P<0.001) as compared with those in the first and second trimesters. However, the changes in myometrial tissues were not statistically significant. Late pregnant uteri were divided into two groups of pregnant women either with or without labor pain. The concentrations of of PGE_1 and PGF_<2α> in myometrial tissues were slightly higher in the labor pain group, but a notable difference was not detected. The results of each steroid hormone administration indicated that estradiol treatment alone markedly (P<0.01) increased of PGE_1 and PGF_<2α> concentration in both myometrial and cervical tissues, whereas progesterone progesterone combined with estradiol treatment did not effect any remarkable changes.
