Relationship between diffraction intensity of the order line and tension development was studied in frog skeletal muscle. (1) The intensity change upon electrical stimulation to the single fiber was similar with the pattern of tension development, although the intensity change preceded the tension development during stimulation. Summation of the optical response was more strikingly observed than that of the tension development. (2) Caffeine (3 mM or 6 mM) in the Ringer's solution caused a decrease in the intensity with a simultaneous development of tension in intact single fibers. The effects of A23187 and dantrolene on Ca^<2+> mobilization from the sarcoplasmic reticulum were evaluated. These results suggest that tension development can be monitored with the magnitude of diffraction intensity change in the first order line.
