STUDIES ON THE ZYMOGRAM OF PEPSINOGENS IN HUMAN GASTRIC MUCOSA

Zymograms of pepsinogens from human gastric mucosa were investigated by using polyacrylamide gel electrophoresis, and the clinical usefulness was evaluated. Gastric mucosal samples were surgically obtained from 5 patients with duodenal ulcers, and 42 samoles of the fundic mucosa and 32 samples of the pyloric mucosa were separately taken by endoscopic biopsy from 42 subjects who were diagnosed as being free of gastroduodenal diseases. 1. All the acid protease bands were divided into three major groups; i.e. fast moving bands, intermediate bands, and one slow moving band. Fast moving and intermediate bands along with one slow moving band were observed in the fundic mucosa, and fast moving bands along with one slow moving band in the pyloric mucosa. Pepsinogens were separatd into 12 bands with the exception of the 13th or slow moving band. 2. Fast moving bands in the fundic mucosa were classified into two separated types (Types A and B), and those in the pyloric mucosa into five types (Types C, D, E, F, and G). Also, fast moving bands in both the fundic and pyloric mucosae were divided into four zymogram patterns (F-1, F-2, F-3 and F-4; P-1, P-2, P-3, and P-4). 3. The incidence of each separated type in the pyloric mucosa appeared to be influenced by atrophic changes. 4.Pepsinogen activity in Type A was affected by atrophic changes in the fundic mucosa. Significant differences of pepsinogen activity were noted between Types E and F in the pyloric mucosa.
