In our previous report, the indirect fluorescent antibody technique was first extended to the serological investigation of antagonistic Streptomyces. Further studies o,n the fundamental problems for its application were carried out with several strains of Streptomyces which produce anti-tumor antibiotics. Using the same procedure for staining as previously described, there observed no marked difference between agglutination titer and that of fluorescent antibody technique. Spore or sporophore of the organism also proved to be satisfactory as an antigen. S. aburaviensis gave orange-red autofluorescence under the fluorescent microscope. However, the autofluorescence was weak and easily distinguishable from a characteristic fluorescing antigen-antibody reaction. Six species which are somewhat related to each other in their cultural characteristics were investigated by this method. No serological interrelationship was found among these species. Employing these antisera of known anti-tumor antibiotic sources, serological identification of fresh isolates was attempted. Antagonistic Streptomyces sp. OK 28-14 was immediately identified as S. pluricolorescens serologically. Two other strains, which also produced anti-tumor substances, were differentiated from any of test species and the products of these strains were found to be different from those of above known species. Even if S. sp. O-2 or S. sp. CK 147-8 produces antibiotic of actinomycin- or quinoxaline-group respectheir serological identification with antiserum of 5. antibioticus (actinomycin A. source) or S. echinatus (echinomycin source) was unsuccessful. It is well known that there are several compounds among Streptomyces antibiotics which possess phenoxazone or quinoxaline skeleton in common but differ in their amino acid composition. The antibiotics of 5. sp. O-2 and S. sp. CK 147-8 are assumed to be different from actinomycin A and echinomycin in their amino acid composition.
