The physiological activities and clinical effects of 3, 5, 3'-1-triiodothyronine (T_3) have been studied in man and animal. The following results were obtained : (1) It was thought that thyroxine (T_4) and T_3 had crossed the placenta in man. The rate of passage of T_3 was more rapidly than T_4. (2) A rapid metabolic turnover rate of T_3 than of T_4 has been demonstrated in rats. The main excretory pathway of T_4 is in the feces and T_3 is in the urine. (3) The rate of disapperance of I^<131> labeled T_3 in man was much more rapid than of T_4. The half-time of I131 labeled T_3 was shorter in hyperthyroid patients and longer in athyrotic cretinism than euthyroid patients. (4) Before and after the oral administration of T_3 for five days, I^<131> thyroidal uptakes were determined in simple goiters and hyperthyroid patients for differential diagnosis. A significant change of uptake was demonstrated in the simple goiters, but no change in the hyperthyroidismus. (5) Hypothyroid children were treated with T_3 for 13 months. It was found to possess qualitatively identical effects of T_3 on clinical status and salts metabolism and balance study as T_4. No difference of the improvement on the height was found between T_3 and T_4. The apperance of ossification centers in the patients treated with T_3 was, however, found, more slowly than with T_4.
