Tetrodotoxin crystals (T. C.) inhibited markedly the contraction of isolated guinea-pig's ileum produced by nicotine and serotonin, while T. C. had no inhibitory effect to the contraction produced by acetylcholine and histamine. The peristaltic action of isolated ileum disappeared by the application of T.C. T.C. did not alter the acetylcholine content in the stretched isolated intestine wall muscle. T. C. had a local anesthetic action. The contraction of the rectus muscle of the frog was inhibited by cocaine or procaine while unaffected by T. C. The relaxant effect of nicotine in the guinea-pig's tracheal chain abolished following pretreatment with hexamethonium, cocaine and T. C. The bronchoconstriction produced by physostigmine and nicotine in the guinea-pig's tracheal chain abolished after hexamethonium, cocaine and T.C. T. C. had no atropinic, antihistaminic or antiadrenaline action in the tracheal chain and unaffected the anaphylactic response of tracheal muscle by egg albumine.
