The methods for the biological assay of 5-hydroxytryptamine (5-HT) were studied on isolated guinea-pig ileum, isolated rat colon and isolated fundus of rat stomach by means of the apparatus of Magnus. The results thus obtained were as follow : 1. The rat fundal strip prepared according to the method of Vane with slight modifi cation was the most sensitive to 5-HT. Usually the preparation responded to 5-HT below 2×10^<-9>, very often to 5-HT 10^<-11>, and sometimes to 5-HT 10^<-12>. 2. The main advantages of the rat fundal strip are that rats of either sex can be used, that it does not need any sre-treatment, for example, starvation or injections of hormones, that individual variation on sensitivity to 5-HT are not observed and that reproducible responses to 5-HT are always exhibited. 3. When 5-HT in the tissues was estimated on the rat fundal strip, the possible interferences by substance P, potassium salts, adenbsine compounds and posterior pituitary hormones were eliminated by using the double extraction of the tissues with acetone ; by adrenaline and noradrenaline eliminated by incubating the extracts with mushroom juice which contains a polyphenoloxidase enzyme; by acetylcholine eliminated by adding atropine to the bathing fluid at this bioassay. There is no interference by histamine, because histamine has to be given in much more doses than 5-HT to produce any effect at all in this preparation.
