Experiments were carried out in vitro to study the role of thiamine in the formation of melanin pigment. It was proved both spectrophotometicaly and manometicaly that a small amount of thiamine and it's diphosphate ester (thiamine diphosphate) added in tyrosinetyrosinase reaction caused definit decreasing oxygen uptake and made a lag period. This fact proved that a reducing mechanisum was formed in tyrosin-tyrosinase reaction in the presence of thiamine and it's derivates. The reducing process thus formed reduce dopaquinone to dopa (←) [figure] Thiamine take part in the formation of melanin in this way reproducing dopa (a catalyst in the reaction of tyrosine to dopa) and being oxidized itself to thiamine disulfide.
