Isoniazid one of the chemotherapeutic drugs for tuberculosis, plays an important role in the treatment of pulmonary tuberculosis. As, however, tubercle bacilli soon acquire resistance to Isoniazid there have been reports from many sources on the decrease of virulence of tubercle bacilli resistant to Isoniazid and on negative catalase reaction tests. The experiments conducted by the author on tubercle bacilli resistant to Isoniazid in guinea pigs and mice produced the following results : 1. The virulence against guinea pigs of tubercle bacilli was markedly reduced as compared with that of tubercle bacilli sensitive to it. 2. When a mixture in epual amount of tubercle bacilli resistant to Isoniazid and those sensitive to it had been injected into guinea pigs, the bacilli retrieved from the internal organs were mainly those sensitive to Isoniazid. 3. The degree of resistance of tubercle bacilli to Isoniazid paralleled the incidence of negative reactions to catalase tests. 4. Tests in which sputa of tuberculous patients were injected into guinea pigs showed that the virulence of tubercle bacilli resistant to more than 10 γ of Isoniazid was reduced. 5. The degree of resistance of tubercle bacilli detected in sputa was compared with that of tubercle bacilli retrieved from animals to which the sputa had been injected. It was shown that the tubercle bacilli retrieved from the site of injection tended to be less resistant to Isoniazid than those detected in sputa, and that those retrieved from the internal organs tended to be less resistant to Isoniazid than those retrieved from the site of injection.
