Stoll's egg counting method has been widely in use for estimation of the number of parasite worm eggs in the feces. In an effort to develop a simpler and more reliable method, the author performed studies on this method, comparing the counts made by different techniques on samples of feces from individuals with Hookworm infestation. The results are summarized as follows : (1) It was shown that a modification of author's which favors the use of 1.5gm. samples was technically more advantageous and no less efficient than the original method using 3gm. samples. (2) It was shown that direct quantitative smear method was as valuable and efficient as Stoll'S method in estimating the number of worm eggs in the feces. Calculation by this method showed that the number of Hookworm eggs in a stool was distributed according to Poisson distribution. (3) Tests performed to compare the virtues of Stoll'd original method, a modification of the author's and smear method by using feces with varying degrees of Hookworm infestation showed that the original method achieved better results with feces containing less than one hundred eggs per gram, the three methods achieved more or less the same results with feces containing one hundred to one thousand eggs per gram and a modification of author's had technical advantage and achieved better results with feces containing over one thousand eggs per gram. (4) Comparison of the counts made one time each on ten 3gm. samples and ten times on one 3gm. sample of the same feces showed that there was no difference in the mean value and distribution of eggs. (5) Determination of water content of feces revealed that the ratio of the volume of dried feces to the total volume of feces was 31 to 27% in formed stools, 26 to 21% in soft formed stools and 20 to 18% in mushy, nearly formed stools. There were significant differences between these samples of feces.(6) There was no marked difference in the water content between the hard and soft portions of a formed stool, nor was there a significant difference in the number of eggs between the two portions. (7) The presence of undigested dusts in a formed digested stool had no influence on the distribution of.eggs in any given samples from it, so that there was no significant difference between these samples. (8) Calculation by Stoll's method revealed that the number of Hookworm eggs in a stool was distributed according to Poisson distribution.
